I believe I've installed DSD+ successfully? I'm using an RSP2 running SDRuno. I am tuned to a frequency that I know is DMR but all I hear when there's a transmission is noise. Listening to the same frequency with an AOR DV-1 the audio is clear. What haven't I done? See attached screenshot.

Assuming you are piping the receiver audio from SDRuno out the VB cable, brettbolf is correct.
dsdplus -i3 -o1 -v4
-i3 says grab input audio from line 3, which is your VB cable
-o1 says play decoded audio on line 1, which is your speakers
-v4 isn't necessary - it's the verbosity level - but I always run -v4
Now, in SDRuno, you need to find the area in SDRuno where you turn DeEmphasis on/off and make sure it is OFF. That would be the DEEM button and should be under the SDRuno EX Control. See screenshot.

Do you have DSD+ decoding anything? In your last screen shot, you had DSD+ listening to your microphone...
You are close, but your audio input and output are not set correctly based on your screen capture in your first post.
In the lower right corner of your capture there are three blocks/paragraphs about audio devices. The first (input) is three lines of what DSD+ found. The second (output) two lines of what DSD+ found. The third is how DSD+ connected by number to your inputs/outputs without being directed by command line switches when you started DSD+.
Now that you know the numbers you need to close DSD+ and restart with command line switches to make the correct connection.
Output from your SDR program must be directed to a VAC cable. That cable is to become the input to DSD+. DSD+ must be connected to the speakers.
Once DSD+ has input from the SDR then the pane in the upper right will no longer be flat lined and should show some ripple noise. When receiving a signal you should see higher amplitude signal looking a little like distorted sine waves. At this point you should be able to decode control channel information which is easier than voice channel for me.
Decoding voice is not like turning on a radio and dialing in the frequency. You are going to have to play with frequency, bandwidth and amplitude. Once you are at that point then look into the fine tuning parameter to improve the decode. Also brush up on your bat file skills to make it easier to start DSD+ with the command switches without a lot of keyboard typing.

@slicerwizard -- I could take a screenshot but the main DSD+ window which shows the information about the soundcards, that window gets filled up with all sorts of details all of which I guess are to do with decoding a DMR signal.

You should be verifying that the DMR signal actually contains voice traffic. The DSD+ event log window should be reporting voice calls. No point worrying about hearing voice if you're not seeing voice...
@slicerwizard -- I'm actually tuned into a known digital (DMR) frequency. The whole load of whatever it is that scrolls up in the main DSD+ window seems full of errors and other seemingly incorrect information -- but that I believe is because I need through learning how to fine tune/tweek things. I need to figure out how to get DSD+ to direct it's audio output to the sound card -- it seems it's hearing audio from SDRuno ok but when I set the VB Cable as my default sound device then no audio whatsoever can be heard.

DMR and DMR voice are not the same thing. So far, you have not stated that it's a known voice channel.
You need to tell SDRuno to use the correct bandwidth (about 8 kHz), the correct demodulation method (FM/Digital) and the correct noise filtering (none)
Based on your initial screen shot, DSD+ would use your speakers by default.
a) You shouldn't ever be hearing audio from SDRuno if you have its audio directed to DSD+
b) There is no good reason to make VB Cable the default sound device
I just got my RSPduo thinking I could replace my two RTL-SDR dongles to follow trunked radio systems. But it ain't working like I thought it would.
My question is ,,, why do you say there is no good reason to make VB Cable the default sound device?
So, how then is audio to get from SDRuno to DSD+ if not by VB Cable? Or, are you saying only there is no need to change the setting of the sound devices so that VB Cable is the default?
And, is it possible to use RSPduo to follow a trunked radio system like the two dongle system?

Because it just messes things up. If you do something else like play a video, the sound goes to VB Cable instead of to your speakers where it belongs.
You tell your SDR software to use VB Cable. Same with DSD+.
Not that I'm aware of.
