I was wondering if two people were hooked up to the same Echolink repeater via their computers, would they be able to talk to each other or not? I made a diagram of what I am trying to explain.

The EchoLink node attached to the repeater needs to be set up to allow multiple connections.
If not then the first person on a PC will be able to connect and the second person will get a "busy" connect rejection.

ok. How would I be able to find out if the repeater allows multiple connections?

If you don't have access to the computer running the EchoLink node then you will never know for certain how it is set up.
You can get several people to try to connect to it at the same time and see what happens.
You can ask the repeater owner as Bill said.

Look at the Conferencing box in the Connections tab in the Preferences menu.
In the link above click on Configuration and Preferences on the menu on right side of the page.

I think most repeaters that have Echolink can have more than one person connected by Echolink. I know the repeaters I have used while checking into nets have had 4-5 people at a time logged in and if we wanted, all of us could talk to each other if we wanted. Echolink also affords the ability to go direct with others on Echolink without going through a repeater. I have done it in the past many times. I believe you can only do one person at a time though if you go direct on Echolink.

there are a lot of options. It depends on how you set it up. Sysop mode and end user mode can both have multiple connections, one connection, a list of allowed stations, a list of banned stations. I have never used the smart phone app so I don't know if it is the same way.
